Fluorescent Lactic Acid Bacteria and Bifidobacteria as Vehicles of DNA Microbial Biosensors

چکیده
Control and quantification of effector molecules such as heavy metals, toxins or other target molecules is of great biotechnological, social and economic interest. Microorganisms have regulatory proteins that recognize and modify the gene expression in the presence or absence of these compounds (effector molecules) by means of binding to gene sequences. The association of these recognizing gene sequences to reporter genes will allow the detection of effector molecules of interest with high sensitivity. Once investigators have these two elements—recognizing gene sequences and reporter genes that emit signals—we need a suitable vehicle to introduce both elements. Here, we suggest lactic acid bacteria (LAB) and bifidobacteria as promising carrier microorganisms for these molecular biosensors. The use of fluorescent proteins as well as food-grade vectors and clustered regularly interspaced short palindromic repeats (CRISPR) are indispensable tools for introducing biosensors into these microorganisms. The use of these LAB and bifidobacteria would be of special interest for studying the intestinal environment or other complex ecosystems. The great variety of species adapted to many environments, as well as the possibility of applying several protocols for their transformation with recognizing gene sequences and reporter genes are considerable advantages. Finally, an effort must be made to find recognizable gene sequences.

Polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) and phylogenetic analysis were used for molecular identification of lactic acid bacteria (LABs) isolated from Apis mellifera. Eighteen honeybee workers were collected from three different apiaries in West Azerbaijan. LABs from the gut of honeybees were isolated and cultured using routine biochemical proce...

Probiotic fermented sausages are safe and healthy meat products. Semi-dry fermented sausages were manufactured from camel meat inoculated with Lactobacillus casei and Lactobacillus paracasei and control. All treatments were analyzed for the physico-chemical characteristics (Protein, Moisture, Fat, Ash, Lactic acid value and pH), microbiological features (total aerobic, total mold and yeast...

The gastrointestinal tract of animals harbors microbial flora. Lactic acid bacteria and bifidobacteria are members in the flora of many animals. The organisms have been reported to have several health-promoting effects on host animals (Nagao et al., 2000; Ouwehand et al., 2002), and thus the flora of the organisms in feces and/or gastrointestinal contents have been actively studied on many anim...
متن کاملThe effects of a microbial inoculant and formic acid as silage additives on chemical composition, ruminal degradability and nutrient digestibility of corn silage in sheep
The effects of a microbial inoculant (containing propionic and lactic acid bacteria) and formic acid onchemical composition, ruminal degradability of dry matter (DM) and nutrient digestibility of corn silage wereexamined. Whole-plant corn was ensiled for 60 days in plastic polyethylene bags, and three treatments werecompared, 1: control (no additive), 2: Propionibacterium acidipropionici plus L...
